Abstract: Ce travail est consacré à l’étude phytochimique et biologique des extraits de la plante Anvillea radiata de la famille d’Asteraceae. Diverses méthodes sont appliquées tel que les techniques chromatographiques de purification et isolation. Trois produits purs sont isolés, deux sesquiterpenes (9α-hydroxyparthenolide, 9β-hydroxyparthenolide) et un acide phénolique (Acide 3,5-O-Dicaffeoylquinic) est isolé pour la première fois dans le genre d’Anvillea. Pour déterminer les structures des produits isolés on a combiné les différentes méthodes d’analyses spectroscopiques telles que la résonance magnétique nucléaire et ces différentes applications (RMN-1H, RMN-13C, DEPT, COSY, HECOOR et NOESY) ainsi que la spectroscopie de masse (ES) et l’IR. L’activité antioxydante est évaluée en utilisant les tests de DPPH, test d’ABTS, blanchissement du β-carotène, et le test du pouvoir réducteur. Dans un premier temps, la teneur en polyphénols totaux et en flavonoïdes est effectuée par la méthode de Folin Cecaltau et AlCl3 respectivement. L’extrait d’acétate d’éthyle a montré une richesse en polyphénols totaux et en flavonoïdes avec des proportions de l’ordre de (600.09 11.0 mg EAG/g d’extrait) et (132.20±1.49 mg EQ/g d’extrait) respectivement. Par ailleurs, cet extrait a montré une très forte activité anti-radicalaire vis-à-vis du radical DPPH avec IC50 égale à (0.0026±0.002 mg/mL). Pour l’ABTS on a enregistré un IC50 égale à (0.001±0.002 mg/mL) même supérieure à celle du BHT. L’extrait a montré un pouvoir inhibiteur important du blanchiment du β-carotène avec un IC50 égale à (0.014±0.002 mg/mL) par rapport au BHT (0.04±0.001 mg/mL). Les deux extraits acétate d’éthyle et butanolique ont montré une meilleure capacité réductrice que l’acide ascorbique (EC50=0.05 mg/mL). L’effet antibactérien des extraits est évalué par le test de diffusion sur l’agar vis-à-vis de cinq souches bactériennes. Les résultats révèlent que l’extrait chloroformique a exercé un effet antibactérien considérable. L’activité antidiabétique de l’Anvillea radiata a révélé un effet important de l’extrait chloroformique et des deux sesquiterpènes lactones de l’inhibition de l’enzyme d’α-glucosidase. L’activité allélopathique a montré que les deux extraits chloroformique et acétate d’éthyle ont un effet inhibiteur sur la germination et la croissance de Radis.
This work is dedicated on the phytochemical and biological study of Anvillea radiata, a medical plant from the Asteraceae family. Many methods have been used, such as chromatography, purification and isolation. Three compounds have been isolated: two sesquiterpenes lactones (9α- hydroxyparthenolide, 9β-hydroxyparthenolide) and one phenolic acid (Acide 3,5-O- Dicaffeoylquinic) is isolated for the first time from specie. To determine the structures of the isolated products, we combined different spectroscopic methods, these include magnetic nuclear resonance, and its different functions (NMR-1H, NMR -13C, DEPT, COSY, HECOOR and NOESY). We also used Mass Spectroscopy (ES) and IR. Antioxidant activity was evaluated using the DPPH, ABTS, β-carotene bleaching, power reducer tests. Firstly, polyphenols and flavonoids content was determined using the Folin Ciocalteu reagent and AlCl3, respectively. Results showed that ethyl acetate extract is rich in polyphenols and flavonoids with values of (600.09 ± 11.0 mg EAG/g) and (132.20±1.49 mg EQ/g) respectively. The extract showed a very strong antiradical activity against DPPH radical, with IC50 values of (0.0026±0.002 mg/mL). ABTS showed an IC50 value of (0.001±0.002 mg/mL) which is much greater than that of BHT. The extract showed a strong inhibiting capacity of β- carotene bleaching with an IC50 values of (0.014±0.002 mg/mL) compared to BHT (0.04±0.001 mg/mL). Ethyl acetate and n-butanol extracts have a better reducing capacity as ascorbic acid (EC50=0.05 mg/mL). Antibacterial effect was evaluated by agar diffusion assay toward five bacterial strains. The results showed that the chloroform extract had a considerable antibacterial effect. Anti-diabetic properties from the Anvillea radiata showed an important effect on the chloroform extract, and 2 lactone sesquiterpene C10 and C11, which inhibits the α- glucosidase enzyme. Allelopathic activity showed that two chloroform and ethyl acetate extracts have an inhibiting effect on the germination and growth of radishes.
