Abstract: Ce travail est conçu à l’étude des polysaccharides hydrosolubles issus de la gomme-résine de Ferula assa-foetida. C’est une plante spontanée de la famille des Apiaceae à caractère médicinal récoltée dans la région de Ghardaïa, située au Sahara septentrional Est algérien. Les polysaccharides sont précipités par acétone, puis lyophilisés. Le rendement massique de l’extrait brut des polysaccharides hydrosolubles de la gomme-résine est de 2,43%. L’étude de la composition chimique de l’extrait polysaccharidiques hydrosolubles, laisse apparaître des taux 77±0,22% en oses totaux, 46,35±0,015% en oses neutres, 18,7±0,045% en oses acides et 7,37±0,023% en protéines. Il apparaît que l’extrait polysaccharidique de la gomme-résine est constitué des hétéro-polysaccharides. Les analyses chromatographiques en utilisant trois systèmes de CCM après hydrolyse par TFA à 2 M durant 4 heures à 100 °C révèlent que les polysaccharides de la gomme-résine sont constitués principalement d’acide galacturonique, d'acide glucuronique, d’arabinose, de galactose, de rhamnose et de xylose. La diversité structurale remarquable des polysaccharides, permet d’espérer un large spectre de propriétés biologiques. L’étude de l’activité antidiabétique des polysaccharides hydrosolubles, porte sur la détermination de leur pouvoir inhibitrice de l’enzyme α-D-glucosidase. L’étude a montré que l’extrait des polysaccharides a un pouvoir inhibiteur de l’α-D-glucosidase important qui est de 77,66% pour une concentration maximale de 100 mg/ml, en comparaison à l’acarbose comme contrôle positif qui a un fort pouvoir inhibiteur de 100% à partir de la concentration 100 mg/ml. Les résultats de ce travail a confirmé l’effet antidiabétique des polysaccharides. Ce qui soutient l'utilisation traditionnelle de Ferula assa-foetida pour prévenir les complications diabétiques. This work devoted to study of the water-soluble
polysaccharides that were extracted from the gum-resin of Ferula assa-foetida. It is a spontaneous plant of the family Apiaceae used in folk medicine harvested in the region of Ghardaïa, located in the northern Algerian Sahara. The polysaccharides are precipitated by acetone and then lyophilized. The mass yield of the crude extract of the water-soluble polysaccharides of the gum-resin is 2.43%. The study of the chemical composition of the water-soluble polysaccharide extract showed 77 ± 0.22% total oses, 46.35 ± 0.015% neutral oses, 18.7 ± 0.045% acidic and 7,37±0,023% protein. It appears that the polysaccharide extract of the gum-resin consists of the heteropolysaccharides. Chromatographic analyzes using four TLC systems after hydrolysis with 2 M TFA for 4 hours at 100°C. reveal that the gum-resin polysaccharides consist mainly of galacturonic acid, glucuronic acid, arabinose, Galactose, rhamnose and xylose. The remarkable structural diversity of the polysaccharides makes it possible to hope for a broad spectrum of biological properties. The study of the anti-diabetic activity of water-soluble polysaccharides relates to the determination of their inhibitory potency of the enzyme α-D-glucosidase. The study showed that the extract of the polysaccharides has an important α-D-glucosidase inhibitory power which is 77.66% for a maximum concentration of 100 mg /ml, compared to acarbose as a positive control which has a strong inhibitory power of 100% from the concentration 100 mg /ml. The results of this work confirmed the anti-diabetic effect of polysaccharides. This supports the traditional use of Ferula assa-foetida to prevent diabetic complications.
