Abstract: Etude d’une bactériocine (type nisine) produite par deux souches de bactéries lactiques (Lactococcus lactis subsp. lactis) isolées à partir du lait des espèces, cameline et caprine : utilisation dans la bioconservation des viandes La présente étude comporte cinq volets d’investigation complémentaires : - isolement d’une souche d'intérêt " Lactococcus lactis subsp. lactis" à partir du lait de deux espèces laitières, cameline et caprine ; - optimisation de ses conditions de culture par modification du milieu de culture , de sa composition ( addition de fructose ou de glucose à différentes proportions ) et de la durée d’incubation ;- production de bactériocine (de type nisine) et étude de son activité antibactérienne à l'égard d'une souche de Staphylococcus aureus ATCC 25923, par des tests d'antagonisme (puits, spots et disques) ; -optimisation des conditions de conservation des souches d'intérêt isolées, par réfrigération et par congélation sans et avec cryoprotecteurs (saccharose, glycérol et DMSO) ; -essais de bio-conservation des viandes, cameline et de poulet par la bactériocine contenue dans les surnageants de culture des souches lactococciques cameline, caprineet bovine (témoin).L’action in situ des bactériocines est parallèlement étudiée par inoculation des produits carnés par les cultures de lactocoques camelins, caprins et bovins. Après isolement sur M17,les souches ont été purifiées par 5 repiquages successifs et identifiées par des tests physico-chimiques et biochimiques. Les essais d’optimisation des conditions de culture ont montré que le M17 avec une période d’incubation égale à 24 heures convenait mieux aux souches camelines et permettent l’obtention d’une meilleure biomasse et ce quelque soit l’ose ajouté au milieu et ses proportions. Pour les souches caprines, c’est plutôt le milieu MRS qui a permis l’obtention d’une meilleure biomasse et ce quelque soit la nature de l’ose (fruc. glc.) ajouté au milieu et ses proportions et quelque soit la durée d’incubation utilisée. Les tests d’antagonisme réalisés avec les surnageants de culture des lactocoques camelin (SNCM) , (SNCP) et bovin (SNB) par la méthode des disques ont donné des zones d’inhibition (ZI) comparables pour le SNCP et le SNCM. Des zones d’inhibition plus petites ont été obtenues avec le surnageant de la culture de Lactococcus lactis subsp. lactis bovin (SNB). La bactériocine contenue dans tous les surnageants est thermostable, résistante à des pH acides et basiques et à la pepsine, ce qui nous a permis de confirmer qu’il s’agit bien d’une bactériocine (type nisine). Par ailleurs, les essais d’optimisation des conditions de conservation par congélation des souches de lactocoques ont montré que l’utilisation du DMSO, celle du glycérol, ou du saccharose comme cryoprotecteur n’améliorait pas significativement la conservation des souches camelines(p>0.05). La congélation (-20°C) sans cryoprotecteurs suffirait pour conserver les souches camelines durant vingt jours. La conservation des souches caprines est par contre meilleure par congélation en présence de saccharose à 12%. Les analyses préliminaires des viandes, cameline et de poulet ont montré des taux de germes supérieurs aux normes réglementaires, notamment ceux des Staphylococcus aureus, espèce contaminante et altérante des viandes aussi bien rouge que blanches. Dans cette partie de l’étude, les surnageants de culture (les bactériocines) et les souches de lactocoques (production de bactériocines in situ) sont utilisées dans un but de bioconservation de la viande de dromadaires et le poulet, deux produits carnés entrant dans les habitudes alimentaires de la population locale. Nous remarquons que les surnageants et les Lc. présentent une activité inhibitrice presque similaire. Ce résultat est de nature à suggérer que la part de la bactériocine produite par l’espèce Lactococcus lactis subsp. lactis dans le système de défense contre les staphylocoques est importante. Concernant la conservation de la viande cameline, l’activité anti- staphylococcique exercée par la bactériocine contenue dans le surnageant caprin ou par les souches de Lc. caprins (production de bactériocine in situ) semble plus importance que celle exercée par les souches de Lc. camelines et leur surnageant. Concernant, la viande de poulet, des constatations similaires à celles relevées dans le cas de la viande rouge ont été enregistrées. D’une manière générale, une augmentation du taux de Staph. est observée durant les deux premiers jours (J0+1) de conservation des produits carnés expérimentaux. Cette augmentation est plus prononcée dans le cas des produits carnés surcontaminés par la souche de Staphylococcus aureusATCC25923, traités par les Lc. ou par leur surnageants. Le phénomène de rebond a été observé au-delà du 3ème jour (J0+2) de conservation dans la majorité des cas. Cela nous conduit à préconiser une limite de la durée de réfrigération des denrées carnées traitées directement ou indirectement par la bactériocine (type nisine).
ملخص دراسة لبكتريوسين (نوع نيسين) المنتجة من طرف سلالتين من البكتيريا اللاكتيكية Lactococcus lactis subsp.lactis معزولة من حليب الإبل والماعزواختبارها في مجال في الحفظ البيولوجي للحوم تهتم هذه الدراسة بخمسة جوانب تكميلية من التحقيق: - عزل سلالة ذات فائدة Lactococcus lactis subsp. lactis" من حليب ، الناقة والماعز - تحسين ظروف زرعهما عن طريق تعديل الوسط المعيشي، تكوينه (إضافة الفركتوز أو الجلوكوز بنسب مختلفة) ووقت الحضانة؛ -إنتاج بكتيريوسين (نوع نيسين) ودراسة نشاطها المضاد ضد سلالة من المكورات العنقودية الذهبية Staphylococcus aureus ATCC 25923، عن طريق اختبارات العداء (الآبار والبقع والأقراص).- تحسين ظروف حفظ هتين السلالتين ، عن طريق التبريد أو التجميد مع اوبدون مواد حافظة(السكروز، الجلسرول و DMSO)؛-محاولة حفظلحوم (الجمال والدجاج )عن طريق استعمال البكتريوسين الموجؤدة في المواد المواد الطافيةsurnageantsزرع بكتيرياLactococcus الابل، المعز والبقر(مستعملة كشاهد). بعد العزل داخل وسطM17 ، تم تنقيةاالسلالات المعزولة عن طريق 5 زراعات تنقية متتالية تم والتعرف على البكتيريا عن طريق الاختبارات الفيزيائية والكيمياء الحيوية. أظهرت تجارب تحسين ظروف زرع أن M17 مع فترة حضانة 24 ساعة كان أكثر ملائمة للسلالات المعزولة من حليب الإبل حيث يسمح هذا الوسط بالحصول على كتلة حيوية أفضل مهما كان تركيز السكريات المضافة إليه. بالنسبة لسلالات المعزولة من حليب الماعز فإن الوسطMRS مكننا من الحصول على أفضل كتلة حيوية مهما كانت طبيعة و نسبة السكريات المضافة إليه. ومهما كان وقت الحضن المستخدم. أعطت اختبارات العداء التي أجريت علىالمواد الطافية لLactococcus المعزولة من حليب النوق و المعز مناطق تثبيط ZI متقاربة أما المعزولة من حليب البقر والماخودة كشاهد مناطق تثبيط أصغر. إن البكتريوسينات الموجودة في جميع المواد الطافيةsurnageantsمقاومة للحرارة ومقاومة للحموضة والبيبسين، مما سمح لنا بالتأكد من انها بكتيريوسين (نوع نيسين). إضافة إلى ذلك، اختبارات تحسين حفظ هذه البكتيريا عن طريق التجميد أظهرت أن استخدام DMSO، الجلسرول، أو السكروز كا cryoprotecteurs لم يحسن بشكل ملحوظ حفظ البكتيريا(p>0.05). المعزولة من حليب النوق(p>0.05). التجميد (-20 درجة مئوية) دون cryoprotecteurs كان كافيا للحفاظ على هذه البكتيريا لمدة عشرين يوما. من ناحية أخرى، حفظ سلالات البكتيريا المعزولة من حليب الماعزكان افضل عن طريق التجميد في وجود 12٪من السكروز. أظهرت التحاليل الأولية للحوم والجمل والدجاج إن معدلات الجراثيم فاقت المعايير التنظيمية،، بما في ذلك المكورات العنقودية الذهبيةالملوثة للحوم الحمراء و البيضاء. وأظهرت التحاليل الأولية للحوم الابل واالدجاج مستويات أعلى من المستويات العادية للجراثيم، بما في ذلك المكورات العنقودية الذهبية، الملوثة للحوم الحمراء والبيضاء. في هذا الجزء من الدراسة، تم استخدام المواد الطافية (البكتريوسينات) وسلالات البكتيري اللبنية (إنتاج البكتريوسين في الموقع) لغرض الحفظ البيولوجي للحوم الإبل والدجاج، اللذان ينتميان للعادات الغذائية للسكان المحليين. تظهر النتائج نشاط مثبط للبكتيريا العنقودية الذهبية متماثل تقريبا بين المواد الطافية و السلالات المعزولة من الحليب. هذه النتيجة تشير إلى أن البكتيريا التي تنتجها Lactococcus lactis subsp. lactis لها دور هام في نظام الدفاع ضد المكورات العنقودية الذهبية.فيما يتعلق بحفظ لحوم الإبل، فإن النشاط المضاد للفيروسات العنقوديةالذهبية الذي تمارسه البكتيريوسين الموجودة قي المواد الطافية للسلالات المعزولة من حليب الماعز اوالسلالات المعزولة من هذا الاخير (إنتاج البكتيريوسين في الموقع) يبدو أكثر أهمية من ذلك الذي تمارسه البكتيريوسين الموجودة قي المواد الطافية للسلالات المعزولة من حليب الإبل و السلالات المعزولة من نفس الحليب. أما بالنسبة للحم الدجاج، فقد تم تسجيل نتائج مماثلة لتلك المتحصل عليها في اللحوم الحمراء. بشكل عام، لوحظ خلال اليومين الأولين (J0 + 1) من الحفظ التجريبي للحوم زيادة في عدد المكورات العنقودية الذهبية.كما لوحظ. ان هذه الزيادة هي أكثر وضوحا في حالة اللحوم الملوثة بسلالة من المكورات العنقودية الذهبيةATCC25923، والمعالجة بالمواد الطافية للسلالات المعزولة من حليب او للسلالات المعزولة من حليب. وقد لوحظت أيضا ظاهرة انتعاش ما بعد اليوم الثالث(J0+2) من الحفظ في معظم الحالات. هذا يقودنا إلى التوصية بالحد من وقت تبريد منتجات اللحوم المعالجة بشكل مباشر أو غير مباشر بالبكتيريوسين (نوع نيسين)
.Abstract Study of a bacteriocin (nisin type) produced by two strains of Lactococcus lactis subsp. lactis isolated from camel and goat milk and application test in meat bioconservation The present study has five complementary aspects of investigation namely: isolation of required strain "Lactococcus lactis subsp. lactis" from the milk of two dairy species (camelina and goat); the optimization of its culture conditions by the modification of the growing environment, of its composition (addition of fructose or glucose with different dosage) and of the time of the incubation; the production of bacteriocin (nisin type) and the study of its antibacterial activity against a Staphylococcus aureus (ATCC 25923) strain by antagonism tests (wells, spots and discs); the optimization of the conserved conditions of isolated strains by refrigeration, freezing with and without cryoprotectants (sucrose, glycerol and DMSO); the bio-preservation tests of camel and chicken meats by contained bacteriocin in the supernatants culture of camel, goat and bovine strains of Lactococcus lactis subsp. lactis (control sample); and the field action of bacteriocins is studied along with the inoculation of meat products of camelins, goats and cattle lactococci cultures. After isolation on M17, the strains were purified by 5 successive subculturesandidentifiedby physicochemical and biochemical tests. The trials of optimization of the growing conditions showed that the M17 with an incubation period equal to 24 hours was better suited to the camel strains and allow obtaining a better biomass whatever the added ose to the environment and its proportions. For the goat strains, it is rather the MRS environment that has made it possible obtaining a better biomass whatever the nature of the ose (fruc., Glc.) added to the environment and its proportions or the incubation time used. The antagonism tests carried out with the supernatants culture of camel (SNCM), (SNCP) and bovine (SNB) lactococcal by the disk method which gave comparable zones of inhibition (ZI) for the SNCP and the SNCM). Smaller zones of inhibition were obtained with the supernatant culture of Lactococcus lactis subsp. lactis bovinewhich is genetically identified (SNB). The contained bacteriocin in all the supernatants is thermostable and resistant at acidic and basic pH and with pepsin, which allowed us to confirm that it is a bacteriocin (nisin type). Moreover, the optimization tests of frozen storage conditions of lactococci strains showed that the use of DMSO, glycerol, or sucrose as a cryoprotectant did not significantly improve the conservation of camel strains (p>0.05). Freezing (-20 ° C) without cryoprotectants would be enough to keep the camel strings for twenty days. On the other hand, the conservation of goat strains is better by freezing in the presence of 12% sucrose. Preliminary analyzes of camel and chicken meats have shown rates of germs higher than the normal levels, including Staphylococcus aureus, and contaminating species of red as well as white meat. In the present part, the supernatants culture (bacteriocins) and Lactococcus strains (on-site production of bacteriocin) are used for the sake of bioconservation of camel and chicken meat products which are both returning in the eating habits of the local population. Concerning the conservation of camel meat, we notice that supernatants and Lc. show nearly a similar inhibitory activity. This result suggests that the part of bacteriocin produced by Lactococcus lactis subsp. lactis species as defense system against staphylococci is important. Concerning the preservation of camel meat, the anti-staphylococcal activity exercised by the bacteriocin contained in the goat supernatant or by the goats’ Lc. strains (on-site production of bacteriocin) seems more important than that exercised by camelines Lc.strains and their supernatant. Regarding chicken meat, similar findings to those found in the case of red meat were recorded. Generally speaking, an increase in Staph. rate is observed during the two first days (J0 + 1) of experimental preservation of meat products. This increase is more pronounced in the case of meat products which are over contaminated with the Staphylococcus aureusATCC25923 strain and treated with Lc. or by their supernatants. The rebound phenomenon was observed beyond the 3rd day (J0 + 2) of conservation in the majority of cases. This leads us to recommend a limit of the refrigeration time of meat products treated directly or indirectly by bacteriocin (nisin type).
