Abstract: Ce travail est conçu à l’étude des polysaccharides hydrosolubles issus de la plante Plantago albicans. C’est une plante spontanée de la famille des Plantagenaceae à caractère médicinal récoltée dans la région de Ghardaïa, située au Sahara septentrional Est algérien. Les polysaccharides sont précipités par l’isopropanol, puis lyophilisés. Le rendement massique de l’extrait brut des polysaccharides hydrosolubles de la plante est de 9%. L’analyse physicochimique donne des valeurs de 37,23±2,34% d’humidité et 27,57±1,09% de cendres. L’étude de la composition chimique de l’extrait polysaccharidiques hydrosolubles, laisse apparaître des taux 21,4±0,04% en oses totaux, 18,69±0,11% en oses neutres, 2,71±0,02% en oses acides, 6,14±0,03% en oses reducteurs et l’extrait est dépourvu de polyphenols (0%). Il apparaît que l’extrait polysaccharidique de Plantago albicans est constitué des hétéro-polysaccharides. Les analyses chromatographiques en utilisant deux systèmes de CCM après hydrolyse par TFA à 2 M durant 4 heures à 100 °C révèlent que les polysaccharides de Plantago albicans sont constitués principalement d’arabinose, galactose, glucose et xylose. La diversité structurale remarquable des polysaccharides, permet d’espérer un large spectre de propriétés biologiques. L’étude de l’activité antidiabétique des polysaccharides hydrosolubles, porte sur la détermination de leur pouvoir inhibitrice de l’enzyme α-D-glucosidase. L’étude a montré que l’extrait des polysaccharides a un pouvoir inhibiteur modéré de l’α-D-glucosidase qui est de 39,66% pour une concentration maximale de 50 mg/ml, en comparaison à l’acarbose comme contrôle positif qui a un fort pouvoir inhibiteur de 100% à partir de la concentration 100 mg/ml.
This work devoted to study of the water-soluble polysaccharides that were extracted from the plant of Plantago albicans. It is a spontaneous plant of the family Plantagenaceae used in folk medicine harvested in the region of Ghardaïa, located in the northern Algerian Sahara. The polysaccharides are precipitated by isopropanol and then lyophilized. The mass yield of the crude extract of the water-soluble polysaccharides of the plant is 9%. The physico-chemical analysis gives values of 37.23 ± 2.34% of moisture and 27.57 ± 1.09% of ash. The study of the composition of the water-soluble polysaccharide extract showed 2.04 ± 0.04% total oses, 18.69 ± 0.11% neutral oses, 2.71 ± 0.02% acidic, 6,14±0,03% reductive oses and the extract is without polyphenols (0%). It appears that the polysaccharide extract of the Plantago albicans consists of the heteropolysaccharides. Chromatographic analyzes using two systems after hydrolysis with 2 M TFA for 4 hours at 100°C. Reveal that the Plantago albicans polysaccharides consist mainly of arabinose, galactose, glucose and xylose. The remarkable structural diversity of the polysaccharides makes it possible to hope for a broad spectrum of biological properties. The study of the anti-diabetic activity of water-soluble polysaccharides relates to the determination of their inhibitory potency of the enzyme α-D-glucosidase. The study showed that the extract of the polysaccharides has a modest α-D-glucosidase inhibitory which is 39,66% for a maximum concentration of 50 mg /ml, compared to acarbose as a positive control which has a strong inhibitory power of 100% from the concentration 100 mg /ml
