Evaluation de l’activité anticoagulante d' exopolysaccharides issus de Dunaliella salina isolées à partir du Chott Ain El-Beida de la cuvette de Ouargla

Abstract

L’objectif de cette étude est l’isolement et identification des microalgues productrices d’exopolysaccharides à partir d’une zone humide (Chott Ain El Beida), ainsi que l’extraction de leurs exopolysaccharides et l’évaluation de leur activité anticoagulante. L’analyse physicochimiques des échantillons d’eau du Chott Ain El Beida a montré que cette dernière est qualifiée neutre et extrêmement salée. La recherche et l'identification phénotypique des micoalgues révèlent une diversité des taxons phytoplanctoniques du Chott Ain El Beida. Trois groupes ont été identifiés (Diatomées, Cyanobactéries, et Chlorophycées). L’isolement par méthode de dilution, nous a permis de purifier deux espèces, Navicula sp.et Dunaleilla sailna. L’activité anticoagulante des extraits exopolysaccharidiques issus de D.salina a été également évaluée in vitro en utilisant les tests, temps de Quick et temps de céphaline activé. Les résultats indiquent que ces exobiopolyméres sont dotés d’une activité anticoagulante relativement importante et que cette activité est dose dépendante.

الھدف من ھذه الدراسة ھو عزل والتعرف على انواع الطحالب الموجودة في الاراضي الرطبة (شط عین البیضاء)، واستخلاص متعدد السكریات الخارجي بالإضافة الى تقییم نشاطھم كمضادات لتخثر الدم. أظھر التحلیل الفیزیوكیمیائي لعینات ماء شط عین البیضاء أنھا تعتبر معتدلة وشدیدة الملوحة. البحث والتعرف عن المظھر الخارجي للطحالب كشف عن وجود مجموعة متنوعة من العوالق النباتیة في شط عین البیضاء. تم تحدید ثلاث مجموعات (الدیاتوم ، البكتیریا الزرقاء والطحالب الخضراء). سمح لنا العزل بطریقة التخفیف بعزل نوعین من الطحالب دونالیلا سلینا ونفیكولا. تم أیضا تقییم النشاط المضاد للتخثر لمستخلصات متعدد السكریات الخارجي المشتقة من دونالیلا سلینا في المختبر باستخدام، الاختبارین زمن البروترومبین والثرومبوبلاستین النشط، تشیر النتائج الى ان ھذه السكریات المتعددة الخارجیة ذات نشاط مضاد للتخثر بنسبة عالیة یرتفع بتزاید التركیز.

The objective of this study is the isolation and identification of exopolysaccharide producing microalgae at the wetland level (Chott Ain El Beida), as well as the extraction of their exopolysaccharides and the evaluation of their anticoagulant activity. The physicochemical analysis of Chott Ain El Beida water samples showed that it is considered neutral and extremely salty. Research and phenotypic identification of micoalgae reveal a diversity of phytoplankton taxa in Chott Ain El Beida. Three groups were identified (Diatoms, Cyanobacteria and Chlorophyceae) .Isolation by dilution method allowed us to purify two species, Navicula sp.and Dunaleilla sailna. The anticoagulant activity of exopolysaccharide extracts derived from D.salina was also evaluated in vitro using the tests, Quick time and activated partial thromboplastin time. The results indicate that these exobiopolymers are endowed with a relatively high anticoagulant activity and that this activity is dose dependent.
