Abstract: | L’étude porte sur les polysaccharides hydrosolubles issus des extraits d’une plante à caractère médicinal dont Salvia
officinalis, récoltée dans la région de l’Oued N’tissa de Ghardaïa, située au Sahara septentrional Est algérien, sont
obtenus par la macération des feuilles broyées de Salvia officinalis dans l'eau distillée puis lyophilisés, leur précipitation
est faite par l’éthanol. Après un séchage à l’air libre le rendement massique d'extraction des polysaccharides
hydrosolubles est de 2.16%.L'étude de la composition de l’extrait brut des polysaccharides hydrosolubles a donné des
valeurs suivantes : oses totaux 21,7%, oses neutres 37,49%. Les analyses chromatographiques en utilisant deux
systèmes de CCM après hydrolyse par TFA à 2 M durant 4 heures à 100 °C révèlent que les polysaccharides sont
constitués de glucose , mannose et l’acide glucuronique. La diversité structurale remarquable des polysaccharides,
permet d’espérer un large spectre de propriétés biologiques. Dans le but d’étudier l’effet immuno-modulateur des
extraits polysaccharidiques, la phagocytose de Candida albicans, levure pathogène opportuniste, par des leucocytes
humaines opsonisées en présence des polysaccharides, est évaluée. L’étude a montré que l’extrait de polysaccahride a
une activité phagocytaire appréciable de 19% pour une concentration de 150μg /ml de l’extrait de polysaccahridique de
Salvia officinalis. Le contrôle positif montre une activité phagocytaire de 16,33 %. Les résultats de ce travail a confirmé
l’effet immuno-modulatrice des polysaccharides en améliorant l’activité phagocytaire des leucocytes .Ce qui peut
expliquer l’utilisation de Salvia officinalis dans la médecine traditionnelle comme antiinflammatoire The study focuses on water-soluble polysaccharides derived from extracts of a plant with an including Salvia officinalis, harvested in the Oued N'tissa region of Ghardaïa, located in the in the Algerian Eastern Sahara, are obtained by maceration of crushed leaves of Salvia officinalis in distilled water and then freeze-dried, their precipitation is made by ethanol. The remarkable structural diversity of the polysaccharides, allows us to expect a wide range of biological properties. In order to study the effect immuno-modulator of polysaccharide extracts, the phagocytosis of Candida albicans, opportunistic pathogenic yeast, by opsonized human leukocytes in the presence of the polysaccharides, is evaluated. The study showed that polysaccahride extract has an activity of phagocyte content of 19% for a concentration of 150μg /ml of the extract of polysaccahridia of Salvia officinalis. Positive control shows phagocytic activity. of 16.33%. The results of this work confirmed the immuno-modulatory effect of the polysaccharides by improving the phagocytic activity of the leucocytes, which may explain the use of Salvia officinalis in traditional medicine as an anti-inflammatory |
